The following are standard methods used in routine analysis:

  1. A 12-well cell culture plate was prepared with 0.5 ml of cell culture medium containing 1 x 10⁴ bovine adrenal vascular endothelial cells (ACE) per well.
  2. The sample to be tested was diluted in PBS containing 0.2% gelatin. Then, 0.5 ml of the diluted sample was added to each well. The same concentration of sample was replaced every other day. A control well received 0.5 ml of cell culture solution. The plates were incubated at 37°C in a 5% CO₂ atmosphere with saturated humidity for 4–5 days.
  3. Cell growth was quantified using the MTT assay, [³H] deoxythymidine incorporation method, or direct cell counting after trypsin digestion. Dose-response curves were plotted, and the highest dilution that resulted in a 50% maximal proliferative response was defined as 1 U of FGF.
